Yes, something need to be put in sympa.conf.
First things first: did you run sympa_wizard.pl after the install? 
That's the script that's supposed to initialize everything.
Second: the parameter defining the alias file for lists is 
sendmail_aliases (whichever MTA you're using) :

Yes, you can edit it through the web interface: go to the list page, 
then go to Admin > Edit list config > List description / home page > 
Click on edit nest to "List description".
