The think you want is possible but the syntax you use is wrong : use
"search" for a named filter not is_subscriber (is_subscriber is
dedicated to lists (may be a list that use ldap include).
In addition, teh scenario seems strange because you require
authentication but
first rule say you accept user without authentication (authentication
method
SMTP is nearly the same as no authentication) .

Heare is my sugestion :

search(l-foro-subscribe.ldap,[sender])      smtp              ->
request_auth
search(l-foro-subscribe.ldap,[sender])      smtp,smime,md5    -> do_it
